Concentration calculations depend on the amount of peptide present in the vial and the volume of solvent added. Lyophilized preparations often contain counterions, salts, or residual water, so the labeled mass may not equal the mass of the peptide itself. This difference can produce a calculated concentration that is higher than the true peptide concentration. Analytical determination of peptide content, rather than reliance on the vial label alone, reduces this source of error. Uncertainty in volume measurement also contributes, especially when small liquid volumes are handled.
Quality records typically include a certificate of analysis, batch number, molecular weight, purity result, and recommended storage conditions. After reconstitution, a laboratory log may record solvent, final volume, date, and storage location. Such documentation supports reproducibility and allows later investigation if a preparation behaves unexpectedly. Stability studies often examine purity and concentration over time under defined temperatures, but results are not universally transferable between peptides or formulations. Open questions remain about how best to predict aggregation for specific sequences and how much analytical testing is sufficient for routine laboratory work.
Quality control for reconstituted peptides includes recording lot number, solvent, date, and storage conditions. Visual inspection checks clarity, color, and particles, while pH measurement verifies the expected solution environment. Concentration is often estimated by ultraviolet absorbance at 280 nm for peptides containing tryptophan or tyrosine, or by high-performance liquid chromatography. Mass spectrometry can confirm molecular identity before reconstitution. Sterility testing is relevant when microbial contamination would invalidate an experiment, though such testing is not routinely performed in every laboratory.
Once a peptide is dissolved, water becomes a medium for hydrolysis, oxidation, and deamidation. Dry powders often tolerate ambient shipping better than liquid solutions, but the exact stability profile depends on sequence and formulation. Refrigerated storage near 2 to 8 degrees Celsius or frozen storage at minus 20 or minus 80 degrees Celsius is common in laboratories. Repeated freeze-thaw cycles can promote aggregation, precipitation, or loss of activity. Dividing a solution into single-use aliquots before freezing can reduce the number of temperature cycles.

Even with all the precautions taken by medical professionals, infection reportedly occurs in up to 13.9% of patients after stabilization of an open fracture, and in about 0.5-2% of patients who receive joint prostheses. To reduce these numbers, the surfaces of the devices used in these procedures have been altered in hopes of preventing the growth of the bacteria that leads to these infections. This has been achieved by coating titanium devices with an antiseptic combination of chlorhexidine and chloroxylenol. This antiseptic combination successfully prevents the growth of the five main organisms that cause medical-related infections, which include Staphylococcus epidermidis, Methicillin-resistant Staphylococcus aureus, Pseudomonas aeruginosa, Escherichia coli and Candida albicans. Peptide-based gel coating with intrinsic antibacterial activity against Methicillin-resistant Staphylococcus aureus, was also shown to inhibit colonization of titanium implants in mice.
The American Society for Pharmacology and Experimental Therapeutics (ASPET) is a scientific society founded in late 1908 by John Jacob Abel of Johns Hopkins University (also the founder of the American Society for Biochemistry and Molecular Biology), with the aim of promoting the growth of pharmacological research. Many society members are researchers in basic and clinical pharmacology who help develop disease-fighting medications and therapeutics. ASPET is one of the constituent societies of the Federation of American Societies for Experimental Biology (FASEB). The society's headquarters are in Rockville, MD. The current president is Michael F. Jarvis.
Aside from the 22 proteinogenic amino acids, many non-proteinogenic amino acids are known. These either are not found in proteins (for example carnitine, GABA, ornithine) or are not produced directly and in isolation by standard cellular machinery. For example, hydroxyproline is synthesized from proline, and selenomethionine is produced by metabolic modification of methionine. Non-proteinogenic amino acids that are found in proteins are formed by post-translational modification. Such modifications can also determine the localization of the protein, e.g., the addition of long hydrophobic groups can cause a protein to bind to a phospholipid membrane. Examples:
Recent discoveries made through use of high-throughput 'omics approaches include the fact that AAV capsids are post-translationally modified (PTM) during production such as acetylation, methylation, phosphorylation, deamidation, O-GlycNAcylation and SUMOylation throughout capsid proteins VP1, VP2 and VP3. These PTMs differ depending on the manufacturing production platform. Another such discovery is the fact that AAV genomes are epigenetically methylated during production. Besides price, these findings might affect expression kinetics, rAAV receptor binding, trafficking, vector immunogenicity, and expression durability.

In December 2013, consignments of horse meat were seized, and arrests were made. 21 people were arrested on Monday, 16 December, in various parts of southern France. This meat was from horses kept on a farm attached to a scientific laboratory and not certified as fit for human consumption. A dealer from Narbonne was helping police with enquiries. Ac-SDKP (Thymosin Beta 4 Fragment (1–4), Goralatide) is an endogenous tetrapeptide derivative that is the N-acetylated derivative of the fragment composed of amino acids 1-4 derived from cleavage of the N-terminus of the signalling factor thymosin beta-4 by the enzyme prolyl oligopeptidase. It is a selective inhibitor of haematopoietic cell proliferation and has antiinflammatory, anti-fibrotic, and pro-angiogenic properties, with particular applications in preventing the development of kidney fibrosis and cardiac fibrosis following injury to these organs. Large AGE proteins unable to enter the Bowman's capsule are capable of binding to receptors on endothelial and mesangial cells and to the mesangial matrix. Activation of RAGE induces production of a variety of cytokines, including TNFβ, which mediates an inhibition of metalloproteinase and increases production of mesangial matrix, leading to glomerulosclerosis and decreasing kidney function in patients with unusually high AGE levels. Peptides and free adducts, the only breakdown products of AGE that are suitable for urinary excretion, are more aggressive than the AGE proteins from which they are derived, and they can perpetuate related pathology in people with diabetes, even after hyperglycemia has been brought under control.

Ultraviolet absorbance is common when the peptide's extinction coefficient is known. Reverse-phase HPLC with calibration standards can also estimate concentration. Amino acid analysis or quantitative mass spectrometry may be used when higher accuracy is needed.
It usually represents the relative peak area of the target peptide compared with all detected peaks under specific chromatographic conditions. It does not measure biological activity or absolute mass. Different methods or wavelengths can give different purity values.
Yes. Mass spectrometry is widely used because the observed mass can be compared with the expected mass. Peptide mapping or sequence analysis may provide additional confirmation. Identity testing does not by itself establish purity or stability.
Storage time varies with peptide sequence, concentration, solvent, and temperature. No single duration applies to all peptides, and a clear solution can still degrade without a visible change.
